What Is Peptide Reconstitution?
Peptide reconstitution is the process of dissolving a lyophilized (freeze-dried) peptide powder into a liquid solvent — typically bacteriostatic water — to create an injectable solution. Lyophilized peptides are shipped in powder form for stability and shelf-life; they must be reconstituted before use in research protocols.
What You Need for Peptide Reconstitution
Before starting, gather the following: USP-grade bacteriostatic water (0.9% benzyl alcohol), your lyophilized peptide vial with stated mg amount, a sterile 1mL insulin syringe, alcohol swabs, and a reconstitution calculator result. Never shake a reconstituted peptide — always swirl gently.
Step-by-Step Reconstitution Protocol
Step 1 — Calculate your volume: Use the Renew Lab Group reconstitution calculator. Enter your peptide amount in mg and your desired concentration (e.g. 2mg/mL). The calculator outputs exactly how many mL of bacteriostatic water to add.
Step 2 — Swab both vials: Wipe the rubber stopper of the bacteriostatic water vial and the peptide vial with a fresh alcohol swab. Allow 60 seconds to air-dry completely before proceeding.
Step 3 — Draw bacteriostatic water: Insert the syringe needle through the center of the BAC water stopper. Withdraw the exact calculated volume slowly. Remove the syringe.
Step 4 — Inject along the glass wall: Insert the needle into the peptide vial at a 45-degree angle. Slowly push the BAC water in by letting it run down the inside wall of the glass — never inject directly onto the peptide cake. This prevents foaming and degradation.
Step 5 — Swirl gently to dissolve: Gently swirl (never shake) the vial between your palms until the powder fully dissolves. The solution should be clear. If cloudiness persists, allow more time — do not shake.
Step 6 — Label and store: Mark the vial with today’s date. Refrigerate at 2–8°C. Discard 28 days after first entry.
How Much Bacteriostatic Water Should I Use?
The volume of bacteriostatic water depends on your desired final concentration. For example: a 5mg peptide vial reconstituted with 2.5mL of bacteriostatic water creates a 2mg/mL solution.
