WFI Standards — Why Starting Water Quality Determines Final Product Quality

The quality of bacteriostatic water starts with the quality of the water itself — before any benzyl alcohol is added, before any filtration occurs. Water for Injection (WFI) is the pharmaceutical industry’s highest-grade water specification, and it is the mandatory starting material for all injectable pharmaceutical water products, including bacteriostatic water.

USP WFI Specifications

USP defines Water for Injection by four key parameters: (1) Conductivity — ≤1.3 µS/cm at 25°C, indicating very low ionic content. (2) Total Organic Carbon (TOC) — ≤500 ppb, indicating very low organic contamination. (3) Endotoxins — ≤0.25 EU/mL, ensuring no bacterial endotoxin burden. (4) Microbial count — ≤10 CFU/100mL for bulk WFI. These specifications collectively ensure a water so pure it is suitable as the base for injectable pharmaceutical products.

How WFI Is Produced

WFI is produced by distillation or by reverse osmosis combined with ultrafiltration. Distillation uses phase change (water becomes steam, is collected and condensed) to separate water from ionic, organic, and microbial contaminants. Membrane-based production (USP-permitted since 2017) uses multiple filtration stages to achieve equivalent purity.

From WFI to Bacteriostatic Water

Once WFI-quality water is confirmed, the production of bacteriostatic water adds one controlled step: pharmaceutical-grade benzyl alcohol at 0.9% concentration. This single addition transforms WFI into a multi-use injectable preservative solution. The entire subsequent process — sterile filtration, cleanroom filling, container closure — is designed to maintain the WFI-grade purity of the starting water throughout.
